    At a Bio-Terrorism Conference at Case Western Reserve University School of Law on March 31, 2006, the government participants were asked what they would do if a superior instructed them not to disclose information to the public about the likely grave health affects of an ongoing bio-terrorist attack. In response, they indicated that they would be reluctant to become a whistleblower. This is not surprising since, despite the federal and state laws that purport to facilitate such whistleblowing for the public good, government whistleblowers routinely have faced loss of promotion, harassment, firing, and in some instances criminal prosecution when they have gone public with their important information. Yet, without government whistleblowers who had the courage to go to the press, the public would never have learned about Watergate, the Iran-Contra scandal, the inhumane practices at Abu Ghraib prison in Iraq, the secret prisons run by the United States in Eastern Europe, or the NSA policy of wiretapping Americans without warrants. These disclosures initiated vital public debate and prompted corrective actions and reforms. The authors argue that the government whistleblower who in good faith discloses information to the press should no longer be treated as an enemy of the state, and provide a legislative proposal to give them a greater degree of protection from retaliation than exists under current legislation

    HDAC inhibitors increase NRF2-signaling in tumour cells and blunt the efficacy of co-adminstered cytotoxic agents

    The NRF2 signalling cascade provides a primary response against electrophilic chemicals and oxidative stress. The activation of NRF2-signaling is anticipated to have adverse clinical consequences; NRF2 is activated in a number of cancers and, additionally, its pharmacological activation by one compound can reduce the toxicity or efficiency of a second agent administered concomitantly. In this work, we have analysed systematically the ability of 152 research, pre-clinical or clinically used drugs to induce an NRF2 response using the MCF7-AREc32 NRF2 reporter. Ten percent of the tested drugs induced an NRF2 response. The NRF2 activators were not restricted to classical cytotoxic alkylating agents but also included a number of emerging anticancer drugs, including an IGF1-R inhibitor (NVP-AEW541), a PIM-1 kinase inhibitor (Pim1 inhibitor 2), a PLK1 inhibitor (BI 2536) and most strikingly seven of nine tested HDAC inhibitors. These findings were further confirmed by demonstrating NRF2-dependent induction of endogenous AKR genes, biomarkers of NRF2 activity. The ability of HDAC inhibitors to stimulate NRF2-signalling did not diminish their own potency as antitumour agents. However, when used to pre-treat cells, they did reduce the efficacy of acrolein. Taken together, our data suggest that the ability of drugs to stimulate NRF2 activity is common and should be investigated as part of the drug-development process

    Modeling Quantum Optical Components, Pulses and Fiber Channels Using OMNeT++

    Quantum Key Distribution (QKD) is an innovative technology which exploits the laws of quantum mechanics to generate and distribute unconditionally secure cryptographic keys. While QKD offers the promise of unconditionally secure key distribution, real world systems are built from non-ideal components which necessitates the need to model and understand the impact these non-idealities have on system performance and security. OMNeT++ has been used as a basis to develop a simulation framework to support this endeavor. This framework, referred to as "qkdX" extends OMNeT++'s module and message abstractions to efficiently model optical components, optical pulses, operating protocols and processes. This paper presents the design of this framework including how OMNeT++'s abstractions have been utilized to model quantum optical components, optical pulses, fiber and free space channels.     Progressing the care, husbandry and management of ageing mice used in scientific studies

    Driven by the longer lifespans of humans, particularly in Westernised societies, and the need to know more about ‘healthy ageing’, ageing mice are being used increasingly in scientific research. Many departments and institutes involved with ageing research have developed their own systems to determine intervention points for potential refinements and to identify humane end points. Several good systems are in use, but variations between them could contribute to poor reproducibility of the science achieved. Working with scientific and regulatory communities in the UK, we have reviewed the clinical signs observed in ageing mice and developed recommendations for enhanced monitoring, behaviour assessment, husbandry and veterinary interventions. We advocate that the default time point for enhanced monitoring should be 15 months of age, unless prior information is available. Importantly, the enhanced monitoring should cause no additional harms to the animals. Where a mouse strain is well characterised, the onset of age-related enhanced monitoring may be modified based on knowledge of the onset of an expected age-related clinical sign. In progeroid models where ageing is accelerated, enhanced monitoring may need to be brought forward. Information on the background strain must be considered, as it influences the onset of age-related clinical signs. The range of ageing models currently used means that there will be no ‘one-size fits all’ solution. Increased awareness of the issues will lead to more refined and consistent husbandry of ageing mice, and application of humane end points will help to reduce the numbers of animals maintained for longer than is scientifically justified

    Experienced physicians benefit from analyzing initial diagnostic hypotheses

    Background: Most incorrect diagnoses involve at least one cognitive error, of which premature closure is the most prevalent. While metacognitive strategies can mitigate premature closure in inexperienced learners, these are rarely studied in experienced physicians. Our objective here was to evaluate the effect of analytic information processing on diagnostic performance of nephrologists and nephrology residents. Methods: We asked nine nephrologists and six nephrology residents at the University of Calgary and Glasgow University to diagnose ten nephrology cases. We provided presenting features along with contextual information, after which we asked for an initial diagnosis. We then primed participants to use either hypothetico-deductive reasoning or scheme-inductive reasoning to analyze the remaining case data and generate a final diagnosis. Results: After analyzing initial hypotheses, both nephrologists and residents improved the accuracy of final diagnoses (31.1% vs. 65.6%, p < 0.001, and 40.0% vs. 70.0%, p < 0.001, respectively). We found a significant interaction between experience and analytic processing strategy (p = 0.002): nephrology residents had significantly increased odds of diagnostic success when using scheme-inductive reasoning (odds ratio [95% confidence interval] 5.69 [1.59, 20.33], p = 0.007), whereas the performance of experienced nephrologists did not differ between strategies (odds ratio 0.57 [0.23, 1.39], p = 0.2). Discussion: Experienced nephrologists and nephrology residents can improve their performance by analyzing initial diagnostic hypotheses. The explanation of the interaction between experience and the effect of different reasoning strategies is unclear, but may relate to preferences in reasoning strategy, or the changes in knowledge structure with experience

    Curriculum innovation:reflection on MSci Integrated Masters with Industrial Experience

    An emerging curriculum innovation within UK Higher Education is MSci Integrated Masters with Industrial Experience. However, we know little about how such programmes may be designed based on feedback and insights from its three main stakeholders: employers, students, and academics. In this paper we reflect on our ongoing work for curriculum development and evaluation, focusing on the MSci Computer Science at Lancaster University. We report on 15 interviews with employers offering placements, MSci students, and academics supervising them. We focus on the critical issue of matching interests and competencies across the three stakeholders to construct industry placements, while allowing for the negotiation of their mutual benefits. Findings also highlight stakeholders’ perceived placement’s challenges and ways of addressing them. We conclude with several suggestions for the MSci curriculum development including mechanisms for managing stakeholders’ expectations, recognising companies’ contributions, strengthening academics’ engagement during placements, and ensuring effective matching process among companies providing placements, students and academics
